Will Graphic Tablets work with the UDK software?

Graphic tablets (In this context) just replaces the mouse. So yeah.
But they are designed to work in a 2d environment, not 3d. So you might not have an easy time.

For just blocking and placing meshes you go better with a mouse but painting terrains is just awesome speedy and percise with a tablet. I recommend it
